Convenient and Fast: PCR purification in only two steps – just bind and elute

Introduction

Cleaning up DNA fragments after enzymatic treatment is a standard routine in most laboratories. Depending on the required downstream process, this includes purification of fragments after methods like PCR, restriction, dephosphorylation, adapter (linker) removal and other applications. In this application note, the performance and results of four different commercial available kits for purification of PCR products are compared. All products are based on the solid-phase extraction using silica Spin Filters. Three of the kits used are based on traditional high-salt chaotropic chemistry covered by Boom patents in the late 1980ies. Analytik Jena’s innuPREP PCRpure Kit is based on the novel, patented Dual Chemistry-Technology (DC-Technology®). This new technology is based on the proprietary combination of chaotropic and anti-chaotropic salts at low concentrations leading to buffers which are ideally adapted to different starting materials. Whilst classical methods require four handling steps at least, the DC-Technology® renders the need of washing and drying steps unnecessary, leading to a rapid Two-Step-Procedure for isolation of PCR products, therefore. Finally all four kits give comparable quality of purified PCR fragments but time to result per sample is halved with the innuPREP PCRpure Kit. Hands-on steps are minimized to an absolute minimum thus also reducing the potential risk of contamination and human errors.
